Laboratory of Dr. Modyanov
was studying different molecular aspects of mammalian Na+K+-
ATPase biology. During my thesis work in his lab I have constructed
the physical map of the cosmid clone containing human gene of Na,K-ATPase
a2
isoform by blot-hybridization analysis with pork Na+K+- ATPase cDNA.
This analysis showed that the cosmid did not contain the complete human
a2
isoform gene. To find clones containing missing 3’-end of a2
gene I have subcloned the most 3’-end BamHI-SalI fragment from the cosmid
clone and used it to screen human genomic library in l
phage. I have characterized seven isolated by cross-hybridization
l
phage clones. I found that two of them contained the missing 3’-end
of the a2 gene. Now this project seems
pitiful, but I remind you: it was 1989 and a lot of things were different
in those days !
Upon my graduation
I have continued working in Modyanov’s lab as Research Associate and used
the similar approach to find the gene for human a1
isoform of Na+K+- ATPase.
